Recovering Cells from Liquid Nitrogen Storage

It is just as important to thaw cells properly as it is to freeze them for storage. It is important to note that LN2 is at 185C. Moving frozen cell vials to dry ice (70C) for shipping or a 80C freezer is a difference of >100C, and thus the cells experience a temperature change on the scale of freezing (0C) to boiling (100C) even though the vial is still frozen! Cell vials that have been removed from LN2 storage for more than a few minutes should be completely thawed, regrown until in log phase, then refrozen.
