Cross-Linking Antibodies to Beads Using Dimethyl Pimelimidate (DMP)

Dimethyl pimelimidate (DMP) (see Fig. 1 in the introductory section Cross-Linking Antibodies to Beads) contains an imidoester at each end of a 7-carbon spacer arm and forms an amidine bond with amino groups at alkaline pH; however, cross-linking is more efficient when performed at pH >8. The amidine bond formation is reversible at very high pH, but is stable at the more neutral pH typically used in an immunoprecipitation (Benashski et al. 1997; Chernukhin and Klenova 2000; Chakrabarti et al. 2003; Vollenkle et al. 2004; Yu et al. 2007; Bons et al. 2008).
