Preparing MBP-Fusion Proteins from Bacteria

Maltose-binding protein (MBP) fusion proteins are commonly used when solubility of the target protein is challenging. Tagging proteins with MBP often enhances the expression levels, solubility, and proper folding of the target protein. MBP-fusion protein purification is mainly based on the strong affinity of MBP, which is a 40-kDa protein (370 residues), to cross-linked amylase. After binding, MBP-fusion proteins can be eluted under very mild physiological conditions enabling the target protein to retain its biological activity.
