Detecting Gold-Labeled Reagents

(Protocol summary only for purposes of this preview site)

Colloidal gold particles bind tightly but not covalently to proteins at pH values that are around the protein's pI. Colloidal gold particles conjugated with a wide range of anti-immunoglobulin antibodies, Protein A, or streptavidin are available commercially. Because some of the bound protein may slowly dissociate from the gold particles, the colloid should be washed before use to remove free protein.
