Preparing Peritoneal Macrophage Feeder Plates

Peritoneal macrophages that are to be used as feeder layer cells should be collected from female animals of the same strain as the immunized animal. They should be collected fresh on the day before or on the day of the fusion. Macrophages provide growth supplements to the establishing hybridomas and will phagocytize low levels of bacterial cells that may have been picked up during the collection of the spleen or lymph node cells from the animal. Macrophages do not typically survive in cell culture for long without special additives, so they will not interfere with the growing hybridoma cells.
